CD/DVD => FOLDER ?

Is it possible, to make that a game would run from a specific folder, not a CD/DVD drive?..

Games don't come in folders, they come in DVDs. The only other possibility is an ISO image of that DVD, which is a single file and not a folder again.

Not having a DVD?

If you mean so you don't need the DVD, you should start by making an ISO of the game disc with an app like ImgBurn to store it in a folder of your liking, once you have made the ISO file open PCSX2 and change your cdvd plugin to "Linuz ISO cdvd" and run PCSX2 normally with "File > Run cd/dvd" and when it asks for the ISO file just point it to the ISO file you made before.

If you meant something else please try to explain better what is it you want to do.
