Try enabling MTVU speedhack and set VU Cycle Stealing to 2.

Config > Emulation Settings > Speedhacks
Alrighty, after doing that it's playable but the sound is still a bit weird and I get some stutter. What settings should I mess with to fix this?
Only a suggestion, because i not plyed yet Shadown of colussos in the emulator, you can try change for use Zerospu2 instead of Spu2 X.

What is your FPS when you get stutter?
in doing that (Destruidor) The sound has gotten worse :/

There's nothing wrong with using SPU2-X

Can you please post screen shots of all your settings?
It drops to 41 then slowly goes back up. that's when the sound gets really bad.
Set Async mix in SPU2-X config. It will keep the sound from slowing down with the FPS

That's exactly why it sounds bad, if your game speed slows down then so does the sound which makes it sound terrible.
